Characteristic of Sexed Sperm with Swim-up Method in Addition of TLR 7/8 Ligand in Dairy Goats

Abstract

Sperm sexing has applications in large-scale farming system and until now flow cytometric is the most effective method of sperm sexing. Nevertheless, this method is expensive, raised time taking and doubts of potential harmful effects on sperm. It is necessary to develop a method of sexing which is cheaper and easier. This study was aims to characterized of sexed sperm that resulted by swim-up method in addition of TLR 7/8 ligand in dairy goats. The research used male dairy goats as a source of semen, and used a completely randomized design with four treatments and six replications. Treatment consists of T0 (without addition of TLR 7/8 ligand), T1 (addition of 0.3 μM TLR 7/8 ligand), T2 (addition of 0.6 μM TLR 7/8 ligand), T3 (addition of 0.9 μM TLR 7/8 ligand) with the parameters of area of ​​the sperm head, X-Y sperm proportion, and quality of X-Y sperm. The result showed that area of sperm head at bottom layer (32,35 μm2) bigger than upper layer (30,39 μm2) at T3. Also, the proportion of X sperm at bottom layer (54,60%) higher than upper layer (47,46%). The sperm motility at bottom layer lower than upper layer. This study showed that at bottom layer was found X sperm more than Y sperm, it seems the role of the ligand. It is concluded that addition of TLR 7/8 ligand can increase the X sperm di bottom layer.
