ESP32-based compact portable device to estimate ram spermatozoa concentration based on the light absorbance

Abstract
Evaluation of ram semen concentration should be carried out as soon as possible after collection because spermatozoa can only survive for a few minutes to 1 hour outside the body. Devices that have been developed for concentration measurement such as spectrophotometer, colorimeter, and hemocytometer tend to be expensive and complex to operate. This invention is a compact portable device to estimate the spermatozoa concentration based on the light absorbance. ESP32 is used to process data from bh1750 sensor (light lux intensity sensor) and ws2812 LED, powered by Li-Ion battery and the results are displayed on an OLED LCD. A total of 15 ram semen samples were evaluated in the laboratory with varying concentration levels. The results were compared with measurements using the developed device with an initial lux of 10538 lux, then the semen samples were placed on a light-proof device to measure the absorbance level with 5 repetitions. The analysis showed a coefficient of determination (R²) of 0.9461, which means that there is a very strong relationship between spermatozoa concentration and light absorbance level, with the smallest value of 30% absorbance (0.05 billion cells/mL) and the largest value of 93% absorbance (1.975 billion cells/mL).
