Observation of the Estrus Cycle in Rats (Rattus novergicus) Pre-pubertal Age, Early Puberty, and Active Reproductive Period

Rats are needed for human purposes in research, therefore humans carry out a breeding system to increase the rat population. Estrouse observation is very important to increase the pregnancy rate in rat. The aim of this study was to evaluate the results of vaginal flushing in three groups of rats (Rattus novergicus) aged 1 month, 3 months and 6 months. Flushing is carried out by setting the micropipette at a volume of 200 µL for three and six month old mice, while for one month old mice it is set at a volume of 100 µL. The flushing liquid is then dripped onto the glass object, then leveled and then dried. After the preparation dries then fixed using methanol for 15 seconds. After that Giemsa staining is carried out for 30 minutes with 5% Giemsa. Result show that one month old rat show an estrus cycle starting from proestrus, estrus, metestrus, and diestrus. However, these phases proceed irregularly. Three month old rat show a more regular estrus cycle than one month old rat. Six month old rat shows a more regular estrus cycle and indicates the mice are ready to reproduce.
